Although it is rare that you can obtain your airline upgrade this way, you will not know unless you ask. However, you CAN ask the ticket agent to add a code to your ticket which will make the gate agent aware that you are eligible for an upgrade. If you know the correct code, this just might work.

Airline competition can work in your favor when trying to get an airline upgrade. This works especially well if you are late due to a competitor's flight not arriving on time. Be sure to tell the ticket agent and you may be awarded that upgrade.
